Sample receiving


1. PURPOSE - This sop explains the procedure for sample receiving for Dengue in virology lab.   

2. STAFF RESPONSIBLE AND AUTHORIZED TO PERFORM THE PROCEDURE: Laboratory technicians

3. PRINCIPLE OF PROCEDURE USED FOR EXAMINATION-Not Applicable

4. PROCEDURE-

TIME & DAYS FOR RECEIVING SAMPLES: Samples are received from patients of civil hospital, Ahmedabad and various district hospitals.

Monday to Friday: 9A.M to 5.00 P.M                                                                                   Saturday: 9-1 P.M

  1. Samples are received at Dengue laboratory, 1st floor, B.J. Medical College.
  2. Blood samples coming from villages, districts are to be brought in vaccine carrier with ice packs to maintain the cold chain.
  3. Samples are received with lab. request form for the test required which mentions name, age, sex, wards no, registration no, and OPD no, address & referring physician. Form also reveals a brief clinical history of the patient and relevant biochemical test results.
  4. Lab request form is checked for the date of onset of symptoms & date of collection of sample by the technician & sample tube is checked for the data like name, age, registration no. and date of collection, quality and quantity of blood/ serum sample.
  5. If any of the information is missing, a repeat form is asked for while sample is retained at the laboratory.
  6. Any leakage is wiped with 1% Na Hypochlorite.
  7. If the interval between onset of symptoms & collection of sample is ≤ than 5 days, sample is tested by dengue rRT PCR or dengue NS1 antigen ELISA and if it is > 5 days, tested by dengue IgM ELISA.
  8. At receiving, each sample is given a lab no. and it is written on sample tube and Lab request form. Time of receiving is entered on LRF. For dengue IgM ELISA,  details of patient are entered in LIS in computer. Refer Laboratory Information System Manual-LSCHABJMC-05.
  9. Samples are preserved in the laboratory for till report dispatch and then discarded by autoclaving.
  1. In case of disparity between sample & LRF, a repeat sample is asked for disparity while first sample is retained and discarded.
  2. LRF are retained in the laboratory for a period of 6 months.

5. PERFORMANCE SPECIFICATION-Not applicable

6. SAMPLE- 5 ml whole blood sample or 1 ml serum sample

7. TYPE OF CONTAINER- Plain test tube or vacuettee or screw capped vial

8. MATERILS, EQUIPMENTS AND REAGENTS USED-
·       Test tube racks
·       Marking pen/pencils
·       Gloves
·       Discarding Beaker

9.  CALIBRATION PROCEDURE – Not applicable

10. QUALITY CONTROL PROCEDURE

CRITERIA FOR REJECTION:
·       Turbid sample
·       Lipaemic sample
·       Hemolysed sample
·       Disparity between lab request form & sample label
      Note: Whenever the sample is inappropriate (as per the above criteria), a repeat sample with lab request form is asked for while the rejected sample is discarded after autoclaving.

16. SAFETY PRECAUTIONS
  • Take precautions while handling the broken sample tubes.
  • Wear gloves while handling the samples.
  • Any blood spill should be treated with 1% Na hypochlorite. ( Refer SOP no 11 )
