Glassware Washing Procedure


1. PURPOSE: This explains the procedure of glassware washing in virology Section.

2. STAFF RESPONSIBLE AND AUTHORIZED TO PERFORM THE PROCEDURE: Laboratory cleaner

3. PRINCIPLE OF PROCEDURE USED FOR EXAMINATION – Not applicable

4. PROCEDURE
PREPARATION OF REAGENTS
Chromic acid
  1. Prepare once or twice a month.
  2. Wear heavy duty full sleeve gloves and eye glasses.
  3. Chill 18 liter water in deep freeze.
  4. Add 18 liter water to 480gm potassium Dichromate powder in tub. Mix it and add 2.5 liter sulfuric acid.
  5. Keep in tub. Cover it. Put the label of date of preparation.
  6. Use for a maximum of 1 month.
Washing soap solution
  1. Add 250 ml Laxbro solution to 10 liter water.
  2. Prepare fresh.

Hydrochloric acid
  1. Take 9 liter distil water. Add 1 liter HCL.
  2. Store in closed container.
  3. Use for a maximum of 15 days.

FLASKS & GLASS BOTTLES:
  1. Immerse used flask in chromic acid for overnight.
  2. Wash under tap water (20 rinses).
  3. Boil in detergent solution for two hours.
  4. Clean it with brush (glassware washing machine can be used for cleaning)
  5. Wash under tap water ( 20 rinses )
  6. Treat with single distilled water for two days in two buckets ( One day in each bucket )
  7. Fifth day dry it in hot air oven.
  8. Plug the glass bottles & flasks, wrap mouth of bottles & flasks with brown paper & sterilize it by hot air oven for two hours at 160°C.

16. SAFETY PRECAUTIONS
  1. Wear Apron. Goggles, heavy duty thick gloves while preparing acid.
  2. Perform all the activities slowly and avoid splashing.
  3. As making acids is hyperthermic reaction, take proper precautions not to burn yourself.
