Preparation of Crystal Violet Stain for staining of cells

1. Scope

This SOP describes the procedure for preparation of 0.025% crystal violet stain in 10% formalin for staining of cells.

2. Definition

3.    Materials:

• Crystal violet
• Formalin
• Distilled water
• Magnet and magnetic stirrer
• 1000 ml Glass bottle
• Spatula
• Weighing balance

4. Procedure

i.       To make 1000 ml stain, take 900 ml distilled water in glass bottle.

ii.     Add 0.25 g crystal violet and 100 ml formalin.

iii.   Stir well on magnetic stirrer.

iv.   Store at room temperature.

5.    Safety conditions

·       Avoid direct contact with reagents.
