1. Scope
- This
procedure explains how to distribute authentic sabin virus reference
strain received from ERC, Mumbai.
From this prepared vials, lab standard of virus are made to check cell
sensitivity.
2. Definition
·
ml =
Milliliter.
·
ART = Aerosol Resistant Tips.
3.
Material
- 5 ampoules of each sabin virus P1, P2, and P3 received from ERC –Bombay on 30/1/2003
·
Pippeters with ART Tips
·
Sterile 1.8ml externally threaded vials
·
Storage box for keeping distributed vials
4.
Procedure
·
All received ampoules were frozen at -200C.
·
Take two ampoules (0.8 ml in each ampoule) of
Authentic Sabin Virus Reference Standard Type 1.
·
Thaw and mix using ART tips and distribute 0.1ml
in each labeled sterile 1.8ml externally threaded vials.
·
Close the cap tightly and store at -200C.
Approximately 16 aliquots are formed.
·
Perform the same steps for Standard Type 2 and
Type 3.
5.
Safety conditions
·
Work only one serotype at a time in biosafety
cabinet.
·
To avoid any leakage tighten the tubes properly.
